Mobile Phase Additives

The chromatographic benefits of employing trifluoroacetic acid (TFA) as the mobile phase additive over formic acid (FA), due to significant reductions in unwanted secondary interactions resulting in peak tailing, has already been discussed for peptide mapping.

The same scenario is present at the protein level, where a vastly superior peak shape is observed in the UV trace with TFA compared with FA. However, once again, the mass spectral signal is significantly more intense when FA is used (Figure 1).
