Tandem mass spectrometry (MS/MS) is a powerful technique for improving quantitative performance, sensitivity, and providing valuable qualitative information in structural determination applications. In this webcast, we explore the principles and practical application of the various modes of MS/MS.
Although we use a triple quadrupole (QQQ) design for our teaching platform, this webcast is applicable to all tandem instrument types, in both GC-MS/MS and LC-MS/MS systems. We explain when to use and how to configure and optimize the common acquisition modes this technique offers.
Using practical examples, we compare EI GC-MS and LC-MS spectra to demonstrate the differences in precursor ion selection, fragmentation, and analytical selectivity. We’ll examine how to optimize precursor ion transmission, select appropriate product ions, and use collision energy to develop effective SRM transitions. The presentation also introduces alternative acquisition approaches, including data-dependent acquisition, parameter-ramped experiments, and pseudo-MS³.
Combining fundamental principles with practical considerations and experimental data, this webcast provides a foundation for understanding, optimizing and developing tandem MS methods across GC-MS/MS and LC-MS/MS applications.
Topics include:
- Understanding QQQ operation, the most common acquisition modes of product ion scanning and SRM
- Autotuning, real time tuning, in-source CID and optimizing precursor ion transmission
- Selecting product ions and optimizing collision energy (Q2)
- Acquisition strategies - product ion scanning, data-dependent acquisition, parameter-ramped experiments and pseudo-MS³
- SRM method development - evaluating transitions for sensitivity and selectivity

Colin Towers (Senior Technical Support Chemist, Crawford Scientific)
After completing a degree in pharmacy, Colin pursued a career in analytical chemistry. Following three years performing routine testing and three years instrument maintenance and calibration, he moved into a role as a method development and validation specialist in contract research. Gaining 11 years’ experience in developing methods for HPLC, LC-MS, GC, GC-MS and SPE in a high throughput commercial environment, this was supplemented by three years lecturing in forensic analytical toxicology and a further three years working as a community pharmacist. Colin has now been with Crawford Scientific since 2011 as a technical specialist, consultant, and trainer.
